Beta-alanine

/ ˈbeɪtə ˈæləniːn / · noun

A non-essential amino acid that combines with histidine in skeletal muscle to form carnosine: an intracellular pH buffer. Supplementation raises muscle carnosine content by 40-80% over 4-12 weeks, improving the muscle's ability to buffer hydrogen ions during high-intensity exercise. Most useful for efforts lasting 30 seconds to 10 minutes.

Mechanism

During glycolytic exercise, hydrogen ion (H+) accumulation drops muscle pH from ~7.0 to ~6.5, inhibiting contractile machinery and creating the burning sensation associated with lactic acidosis. Carnosine accepts H+ ions, slowing the pH drop and extending the time to fatigue. Higher muscle carnosine = more buffering capacity.

Dosing

  • Standard: 4-6 g/day, divided into 2-4 doses to minimize tingling
  • Loading time: 4-12 weeks of consistent dosing to fully saturate muscle carnosine
  • Maintenance: after loading, 1.2-3.0 g/day maintains elevated carnosine
  • Form: sustained-release versions (e.g., CarnoSyn SR) reduce the tingling at higher doses

The tingling (paresthesia)

Doses above 800-1,000 mg in a single sitting cause harmless skin tingling in most users, especially on the face, scalp, and hands. The sensation lasts 30-90 minutes. Splitting doses or using sustained-release formulas eliminates the effect. The tingling is not an indicator of effectiveness: chronic loading works regardless.

Effects (well-established)

  • 1-4 minute efforts: a small but consistent improvement on time-to-exhaustion or repeat-sprint tests, per pooled meta-analysis data (Hobson, Saunders et al., 2012)
  • 30 second to 10 minute efforts: consistent ergogenic benefit
  • HYROX-relevant: stations like the wall ball (~3-4 min), burpee broad jump (~2-4 min), and sandbag lunge (~1-3 min) sit precisely in beta-alanine's effective window

Effects (limited or none)

  • Pure strength events (1RM): no measurable effect
  • Endurance events >25 minutes: minimal effect: fatigue mechanisms shift to substrate availability and central regulation
  • Less than 30 seconds (Phosphocreatine-dominant): creatine is the relevant supplement, not beta-alanine

Safety

  • Well-tolerated; no significant adverse effects at recommended doses (per ISSN position stand)
  • Tingling is harmless
  • Not appropriate for individuals with carnosine-related rare metabolic conditions (rare)

For hybrid athletes

Beta-alanine pairs well with creatine: the two supplements target different energy systems and don't compete. Most elite HYROX athletes' supplement stacks include both. The 4-12 week loading period means beta-alanine should be started in the base phase, not in the final 4 weeks before a goal race.
